Abstract The extracts of Plantain were investigated in the mice model of hyperuricemia. Orthogonal test design was utilized to optimize the extraction conditions including the concentration of ethanol, extraction duration per time, extraction times and amount of ethanol when plantamajoside was selected as the evaluation index. Both plantamajoside and serum uric acid levels were measured by using HPLC. The results indicated that the extracts of Plantain could significantly decrease the serum uric acid level of the hyperuricemic mice (P<0.05). All of the concentration of ethanol, extraction duration per time, extraction times and amount of ethanol had significant effects on the content of plantamajoside extracted from Plantain. The optimum extraction process was to extract two times using tenfold 60% ethanol and 2 h per time
